I believe that Mullis was either confused or most likely regretted what happened with his invention.
In his first paper titled "Specific Enzymatic Amplification of DNA In Vitro: The Polymerase Chain Reaction" (1986) where he describes his novel method of enzymatic amplification he does not mention that this could be used for diagnostic purposes but for detection of allelic variation in gene expression.
However in his next paper titled "Identification of Human Immunodeficiency Virus Sequences by Using In Vitro Enzymatic Amplification and Oligomer Cleavage Detection" (1987) he (alongside his co-authors) mention how his new method (PCR) can identify HIV sequences. Clearly, the identification of something will result in diagnosis of that something.
He obviously got regrets and cold feet when he realised how his method was used by virologists to run wild. Hence why he went public on that infamous video in 2019 to say that his method should not be used for diagnosis. Which actually contradicts his own writing!
This is the danger nature of scientists. Once they realise what they have done in the pursuit of fame and money they cannot reverse what TPTB will do with their produce. They are the real useful idiots.
