Yes, they do. Some maybe all are abrasive and tear the delicate tissues then the contaminants they carry enter the bloodstream. it's a roundabout way to get things into your system.
If it's as easy to transmit covid as they claim, then one wouldn't have to go probe deep into the sinuses. The virus should be all over your respiratory system and a throat swab would catch it.
In my opinion, the deeply inserted nasal swab was a precursor to the anal swab. I believe those test methods were only intended to humiliate people so they will accept others have "authority" over them. Milgram's experiment shows us what can happen next
Anal is actually a very effective ROA, lots of blood vessel.
Back in the day they would administer morphine in the ass because it was more effective. They would rub a wadding full of opiate to the inner wall of your anus.
Addict still do this nowadays its called boofing. Its the best ROA just after intraveinous.

That explains why my daughter uses so many Q-tips when she visits. She's a junky
They use plastic syringe with some water to push it in now, no need for q-tip to boof.
The q-tip are probably for filtering when filling the syringe for IV.
If/When she want to quit try to get her some cyclobenzaprine, it really do wonder for the withdrawal(even for fent).
If she has illness i really wish she heal and get better.
meth suppositories were an option for USAF bomber crews in WWII.
